Alkylating agents have been studied extensively both for their biological effects and for their mode of action. There have been over the past 25 years a veritable deluge of reviews, often focusing on a single aspect or agent or adduct. It can be easily noted that certain analytical techniques were widely used. These were either neutral heating of DNA which liberates 3and 7alkyl purines and/or heating in 0.1 N HC1 which depurinated the other purines, including O'-alkyl G,2 a relative newcomer to the analyst.
